اثر تنظیم کننده پیکس بر پارامترهای رشد آنزیم‌های آنتی اکسیدانی و شاخص‌های اکسیدانی دانه رست دو رقم ساحل و شایان گیاه رست پنبه (Gossypium spp.) در سطوح مختلف خشکی

چکیده

مپی کوات کلرید (پیکس) به عنوان یک ابزار مهم مدیریتی برای کاهش آسیب تنش در گیاه پنبه در مرحله جوانه‌زنی می‌باشد. در این تحقیق، اثر سطوح مختلف خشکی و میزان مپی کوات کلراید (پیکس) بر شاخص‌های رشد و آنزیم‌های آنتی اکسیدانی و نیز میزان مالون دی آلدهید و پراکسید هیدروژن دانه رست دو رقم پنبه تحت شرایط آزمایشگاهی بررسی شد. این تحقیق در یک طرح کاملاً تصادفی با استفاده از غلظت‌های 0 ، 5 و 10 گرم بر لیتر پیکس و نیز اعمال سطوح خشکی با کاربرد پلی اتیلن گلیکول6000 در مقادیر 0، 4- و 8- بار در ظروف پتری بر دو رقم ساحل (حساس به خشکی) و شایان (مقاومت به خشکی) گیاه پنبه انجام شد. شاخص‌های جوانه زنی و نیز پارامترهای فیزیولوژیکی دانه رست‌های تحت تیمار خشکی و پیکس پس از 4 روز اندازه گیری شد. نتایج نشان داد که استفاده از پیکس باعث افزایش درصد جوانه زنی ، طول ریشه‌چـه، وزن خشک و تردانه رست و فعالیت آنزیم‌های پراکسیداز و کاتالاز ارقام پنبه در شرایط تنش خشکی شد در حالیکه اعمال تنش خشکی به تنهایی موجب کاهش پارامترهای ذکر شده گشت. همچنین اعمال خشکی موجب افزایش شاخص‌های اکسیدانی یعنی مالون دی آلدهید و پراکسید هیدروژن در دانه رست‌های دو رقم ساحل و شایان شد در حالی که کاربرد پیکس شاخص‌های نامبرده را کاهش داد. به‌طورکلی نتایج این تحقیق نشان داد کاربرد پیکس سبب بهبود پاسخ‌های رشدی و سیستم آنتی‌اکسیدانی دو رقم پنبه گشت که در این بین پاسخ رقم شایان مناسب‌تر از رقم ساحل بود.

چکیده [English]

Mepiquat chloride (PIX) is an important management tool to reduce stress damage in cotton plants during germination. In this study, the effects of different drought levels and the amount of mepiquat chloride (PIX) were investigated on growth indices and antioxidant enzymes as well as the malondialdehyde and hydrogen peroxide contents of seedlings in two cotton cultivars under laboratory conditions. The study was laid in a completely randomized design using 0, 5, and 10 g/l Pix and also under different drought levels using 0, -4, and -8 bar polyethylene glycol 6000 in petri dishes on two cultivars of cotton plant, namely Sahel (drought sensitive) and Shayan (drought resistant). Germination indices as well as physiological parameters of seedlings were measured under drought and Pix treatment after 4 days. Results showed that the use of Pix increased the germination percentage, root length, dry and fresh weights of seedlings, and the activity of peroxidase and catalase enzymes of cotton cultivars under drought stress while drought stress alone reduced these parameters. Drought also increased oxidative indices such as malondialdehyde and hydrogen peroxide in seedlings of Sahel and Shayan cultivars while Pix application reduced these indices. In general, the results of this study showed that the use of Pix improved growth responses and antioxidant system of the two cotton cultivars and the response of cultivar Shayan was better than Sahel.
